The Cardiac Action of Guanidine


Abstract
There is a limited amount of information on the effects of guanidine on the myocardium and conflicting results have been reported regarding the mechanism of its action. The present studies were attempted to re-examine the nature of the action on the heart.
Results
1. A study of various concentrations of guanidine on the isolated toad¢¥s heart showed that a concentration of 1:10,000 had little or no effects. A concentration of 1:5, 000 or greater exerted a significant cardiostimulant action.
2. The pretreatment with guanidine affected neither the cardiac inhibitory action of acetylcholine nor the cardiac stimulatory action of adrenaline on the isolated toad¢¥s heart.
3. Nicotine produced an initial inhibitory effect followed by a stimulatory effect on the isolated toad¢¥s heart. The prior administration of guanidine blocked only the cardiac inhibitory effect of nicotine.
4. The electrical stimulation of vagosy mpathetic nerve trunk prepared according to the method of Ma*umura resulted in an initial vagal inhibitory action followed by the sympathetic stimulatory action: After the pretreatment with guanidine the stimulation of the nerve trunk produced only the sympathetic cardiostimulant action.
From the above results it may be concluded that guanidine may block the vagal gar_olia in the heart.
